Abstract Glioma is one of the most malignant brain tumor with high rates of recurrence and a poor median survival rate within 2 years of diagnosis. Treatment of glioma is mainly depended on surgery, radiotherapy and chemotherapy. The blood-brain barrier (BBB) as naturally protective layer of brain prevents most of imaging probe and therapeutic drugs from entering the brain tissue, leading to decreased therapeutic efficacy. Focused ultrasound combined with microbubble as a non-invasive technique can temporarily open BBB at a fixed site, revealing attractive application prospects for glioma therapy. Recently, Hynynen et al. finished a first clinical trials of BBB opening for drug delivery in a patient at Canada, further demonstrating the biosafty and effectiveness of ultrasound-opening BBB. In this talk, I will present the current progresses on BBB opening, and discuss the ultrasound nanobiotechnology for crossing the blood–brain barrier for glioma precision therapy.
