Abstract | The cell cycle is a tightly regulated sequential process, and the progression of cell cycle is controlled by a cascade of protein phosphorylation and proteolysis events. Errors in these processes can lead to uncontrolled cell proliferation or chromosomal instability, which are the hallmarks of many diseases, including cancer. VCP/p97, an AAA-ATPase, participates in a wide variety of cellular processes, including cell cycle, and is reported to be dysregulated or overexpressed in many types of cancers or other human diseases. Here I will present two stories on our efforts to dissect the role and molecular mechanisms of VCP mediated cell cycle regulation: (1) VCP/p97 targets the nuclear export and degradation of p27Kip1 during G1 to S phase transition in human breast cancer cells; (2) Polo like kinase 1 (Plk1) phosphorylates VCP/p97 to regulate centrosome maturation during M phase. Our study suggests that VCP is a potential therapeutic or diagnostic target for cancer. |
