Abstract Deregulated NFkB signaling and inflammation are hallmarks of many human ailments. In the past 25 years many proteins which regulate NFkB signaling and hence inflammation have been characterized and mutations in genes coding some of these proteins have been identified as drivers of human diseases. Emerging evidence strongly suggests that mutations in regions of genome that do not code for proteins are also causally linked to human aliments with deregulated NFkB activity and inflammation. A possible interpretation of these observations is that non-coding RNAs (ncRNAs) coded by these regions by themselves may affect NFkB signaling, inflammation and hence pathology. I will present genome scale identification of ncRNAs that regulate NFkB driven inflammation and provide physiological evidence for their involvement in regulating NFkB biology and inflammation in health and disease.
