Talk title  (Pro-)Fluorescent Substrates for Oxidizing and Conjugating Enzymes of Drug Metabolism

Abstract To understand the disposition of drugs and other xenobiotics in the body, it is necessary to measure enzymes transforming their structure. In this presentation it will be presented, how coumarin derivatives are used to measure four different types of drug metabolizing enzymes. CYP enzymes oxidize pro-fluorescent coumarin derivative to fluorescent metabolite. Glucuronosyltransferases and sulfotransferases conjugate fluorescent 7-hydroxycoumarin derivative to non-fluorescent conjugates. Catechol O-methyltransferases methylate weakly fluorescent esculetin to strongly fluorescent scopoletin. The assays can be used to study different types of samples, enzyme kinetics, inhibition of enzymes and to demonstrate function of enzymes for students.
